Intrinsically disordered proteins (IDPs) are proteins that lack a fixed structure and are unstable in cytoplasm when existing alone. But once they bind with target molecules, they adopt a particular three-dimensional structure and exert their functions. We have identified the existence of a complex that has several effects in different time phases during cell division cycles, and reported that one of the components of the complex is an IDP. When its expression is inhibited, apoptosis is induced and it exerts anti-cancer effects.
Development of chemical compounds that inhibit peculiar protein-protein interactions of IDPs has been difficult and no drug has been developed yet. We aim to develop anti-cancer drugs taking a drug discovery approach targeting the IDPs, through a design using helix-loop-helix peptide and Structure Based Drug Design. Our drug discovery research areas also include intestinal flora, arteriosclerosis with age, and iron metabolism.
